🔰Now Culture media দিয়ে আমরা কি করি, এর importance কি?

🔰এখন আসি এদের classification এঃ
অনেক ভাবেই আমরা media গুলোকে classify করতে পারি:
✅ On the basis of consistency :
Solid,Liquid,semisolid আবার
✅ on the basis of composition
Simple/basal & special
✅আবার special media র মধ্যে আরও আছে
💥enriched ,
💥 selective,
💥indicator,
💥transport,
💥storage.
⏹এখন এই media গুলোর প্রত্যেকটিরই কিছু নির্দিষ্ট বৈশিষ্ট্য আছে।
⏺যেমন consistency এর উপর basis করে ভাগ করা liquid media র কিছু disadvantage আছে যেমন
▶ It may not exhibit specific chracteristics
▶Different type of bacteria এখানে অলাদা করা যায় না সহজে।
⏹অন্য দিকেsolid mediaর এই ক্ষেত্রে কিছু advantages আছে যেমনঃ
▶growth is discrete তাই এখানে
▶colony morphology খুব ভালো ভাবে বুঝা যায়
▶ different type of bacteria population এখানে সহজে আলাদা করা যায়
▶shows various characteristics as haemolysis
Pigmentation, so identification becomes easy.
🔰media র জন্য কিছু solidifying agent আছে সেগুলো কি কি?
⏹solidifying agent:
💥Agar
💥Egg white
💥Tomato powder
💥Potato
💥Gelatin.
💥starch
⏹Agar সব চেয়ে ভালো এবং mostly used solidifying agent cause এটার melting point অনেক বেশি এবং এটা Biologically inert অর্থাৎ কোন bacteriar growth promot বা inhibit করবে না।
♐remaining semisolid media is used for transport and to see the motility of organisms
🔰এবার আসি composition based media গুলোর দিকে
🔆 Simple media কে basal media ও বলে কারণ এটা হচ্ছে অন্যান্য অনেক মিডিয়ার base.অর্থাৎ এটার থেকে অন্য media তৈরী করা হয়। e.g.nutrient broth,
Nutrient agar,peptone water
🔆Enriched media:কোন media তে যখন blood,serum, egg ইত্যাদি কিছু ব্যবহার হয় that is called enriched media.eg.blood agar,lowenstein-jensen medium
🔆selective media:এখন কোন media যদি কোন particular bacteriar growth favour করে অর্থাৎ সে bacteriar জন্য selective হয় তবে that is called selective bacteria e.g.Mac Conkey's agar,blood tellurite agar
🔆Indicator media:that means the media contains a indicator which changes when a specific bacteria grows in that,e.g.
Blood agar,Mac Conkeys agar.
⏹এখন আমরা এতক্ষন যা দেখলাম এগুলোর উপর basis করে কিছু media সম্পর্কে শিখবো।এবং দেখবো কিভাবে একটি media কে identify করা যায়।

💥Blood agar media
✅Solid cause agar আছে
✅enriched কারণ blood আছে
✅indicator cause এখানে haemolysis হয়।e.g:beta haemolytic bacteria(strep.pyogens) complete haemolysis করে।
▶এখন এখানে একটা প্রশ্ন আসতে পারে এই media তৈরীতে কিসের blood use হয়?

এর পর
💥lowenstein jensen media, this is
✅Solid cause egg is used
✅Enriched কারণ এটায় egg use হয়।
✅আবার এটা selective, কারণ তিনটা
1⃣M.tuberculosis এর জন্য specific
2⃣malachite green use করা হয় যেটি mycobacterium ছাড়া অন্যান্য bacteria র growth inhibition করে।এবং এটার কারনেই media টির এত সুন্দর একটি color হয়।
3⃣glycerol যেটা M.tuberculosis এরgrowth promote করে কিন্তু M.bovisএর growth promote করে না।সব মিলিয়ে এরা media টি কে M.tuberculosis এর জন্য selective করে
⏹একটা জিনিস খেয়াল রাখতে হবে এইটায় একটা highly contaminant organism culture করা হয় তাই এটাকে normal patridish এ রাখে না।dehydration prevent করাও এখানে আরেকটা উদ্দেশ্য।
⏹another things is খেয়াল করলে দেখা যাবে এটা slopped cause তাতে surface area বাড়ে এবং more oxygen supply হয়ে growth promote করে
💥now Chocolate Agar
✅Solid
✅Selective for fastidious organisms lilke H.influenzae,N.gonorrhoeae,N.meningitidis,s.pneumoniae
✅Enriched.
⏹এই mediaকে আমরা 75-80°c temperature এ 15-20 mint heat দেই।এই heat দিলে RBC lysis হয় এবংmore nutrients release হয়ে media enriched হয়।এবং chocolate color টা হয়।সেই সাথে কিছু toxic products ও destroy হয়।
💥Nutrient agar
✅which is a basal media and it is
Solid and selective
✅composed of nutrient broth+2% agar

1.Mac conkey's agar media
2. XLD(xylose lysine deoxycholate)
3.DCA(deoxycholate citrate agar)
4.tetrathionate broth(selenite F broth)

--tryptic soya broth(tryptone soya,trypticase soya)
--Diphasic tryptone soya
--bile salt broth with or without gention violet
--thipglycolate broth for growth of anaerobes
আর যদি বলে sub culture-
1.blood agar media
2.Mac Conkey's agar media
3.chocolate agar media
এরপর আসি blood culture এর procedure :
1.traditional method-usinf liquid/biphasic media
2.Lytic method-After processing in lytic solution in solid media
3.automated -very rapid(within 6 hour)
