Khwaja Nazimuddin was a child of the Nawab family of Dhaka. He studied at Aligarh, London and Cambridge.

Then he entered politics in 1922. He was the chairman of Dhaka Municipality from 1922-29. He was a member of the Bengal Executive Council from 1937 to 1947.He was the Home Minister of Fazlul Haque's cabinet from 1937 to 1941. He led the Muslim League Parliamentary Party in the Bengal Legislative Assembly. He was a member of the working committee of the All Bengal Muslim League. After partition in 1947, he became the first Chief Minister of East Bengal(1947-48), then the Governor General of Pakistan(1948-51), then the Prime Minister(1951-1953. He played a controversial role in the language movement of East Bengal. He abstained from politics from 1953 to 1962after being ousted by Governor General Golam Mohammad.

He became the president of the newly formed Council Muslim League in 1962 and held the post till his death(1964). From 1962 to 1964, he started an anti-Ayub movement. He had a strong role in the formation of the COP(Combined Opposition Party).
